Under the direction of the PI,the Research Assistant provides technical support for translational and basicresearch in Pediatric Obesity and metabolism in the laboratory in theDepartment of Pediatrics, Le Bonheur Children's Foundation Research Institute,and the University of Tennessee Health Science Center. This positionprovides technical support for ongoing mouse studies (metabolism and bariatricsurgery mouse models) and assists with daily maintenance in the Gnotobioticmouse Facility. This position performs a variety of research experiments at thebench, including histology, DNA isolation, PCR, western blots, and microbialculturing. The Research Assistant performs activities related to researchprojects (design, execution, analysis, and interpretation), data analysis andpresentation, and lab management. The position manages other student andvolunteer activities.
DUTIES AND RESPONSIBILITIES:
Assists with daily operations in the UTHSC Gnotobiotic Facility.
Performs experiments using germ free animals. Sterilizes food, water and bedding. Conduct transfer of germ free animals.
Performs maintenance and repairs to isolators as needed.
Conducts research projects, and help support projects in the laboratory, and assists in training laboratory personnel including students and volunteers.
Performs enzyme linked immunosorbent assays (ELISA). Performs molecular biology techniques, including Western blotting, flow cytometry, and immunohistochemistry. Extract RNA from tissues, perform cDNA synthesis, and conduct expression studies, using quantitative real-time polymerase chain reaction (qPCR). Performs bacterial culture experiments and maintain the anaerobic chamber for screening of the gnotobiotic facility.
Assists in manuscript preparations and grant writing proposals.
Guides, trains, and advises researchers and students on techniques, methods and procedures.
Performs metabolic and food intake analysis of mice models via CLAMS system, and echo MRI. Supports mouse studies focused on metabolism (bariatric surgery, bile diversion, parenteral nutrition in conventional housed mice.
Maintains updated records of mice status/experimental data within in-house mouse database.
Maintains efficient laboratory operation. Keeps orderly up to date laboratory records. Monitors lab inventory and orders supplies as needed. Maintains laboratory or departmental equipment as required.
Maintains proper knowledge and execution of safety procedures.
Performs other duties as assigned or directed in order to meet the goals and objectives of the laboratory and department.
